RNA-sequencing on thyroid samples from fetuses with Down syndrome and fetuses with no genetic/developmental abnormality

The current data pertains RNA-sequencing reads obtained from thyroid samples acquired from fetuses with Down syndrome and fetuses with no genetic/developmental abnormality. Total RNA was isolated from left lobe from thyroid samples using a hand-held homogenizer and the Promega ReliaPrep RNA Miniprep System (Thermo Fisher Scientific). RNA yield was determined with the NanoDrop Microvolume Spectrophotometer (Thermo Fisher Scientific). Fragmentation and mRNA library preparation was performed using the Kapa mRNA Hyperprep Kit (Roche, Basel, Switzerland). Libraries were equimolar pooled and quality was checked on a TapeStation system using the DNA1000 ScreenTape (Agilent Technologies, Santa Clara, CA, USA). Libraries were sequenced with poly(A) selection to sequence all messenger RNA for gene expression analysis on the NovaSeq6000 PE150 (Illumina, San Diego, CA, USA), producing at least 40M 150-bp paired-end reads per library.

Data Access Policy for the study "Multi-omic analysis of thyroid dysfunction in Down Syndrome"

Access requests undergo a comprehensive review by the Data Access Committee (DAC) for the study "Multi-omic analysis of thyroid dysfunction in Down Syndrome" as performed at the Amsterdam UMC. Request will only be granted to qualified users for appropriate use. Applicants are required to submit detailed proposals outlining the purpose, methodology, and ethical considerations of their research. Procedure: 1) The Applicant sends a data access request through European Genome-phenome Archive (EGA) to the DAC. 2) Upon submission, a representative of the DAC will send a Data Request Form (DRF) to the Application. Upon completing the DRF, this form must be returned to the DAC whereupon the evaluation process will commence. 3) When a request is positively evaluated, the applicant will be asked to sign the Data Sharing Agreement. Review and Conditions: - Each DRF is assessed based on the scientific profile of the applicants, the scientific merit, ethical implications, data security plans, and compliance with the relevant regulations, within a specified timeframe. - The DAC reserves the right to conduct periodic reviews to ensure responsible data use. - Approval is contingent on meeting the criteria and compliance with relevant regulations.

Studies are experimental investigations of a particular phenomenon, e.g., case-control studies on a particular trait or cancer research projects reporting matching cancer normal genomes from patients.
